The directions of one way roads in Cities Skylines 2 is based either on the direction of your mouse or the direction of the road you're connecting it to. So, if you have a one way road going to the right, you can't make it suddenly one way to the left without creating a break in the street. You may be able to technically lay it down, but the road will not function properly.

But if you just want to change directions from a single road laid down, just delete the construction and move the mouse in the other direction while laying down the street. This will create a one-way street in a new direction.

One way roads can be spotted all across the road types, with there being one-way roads for small, medium, and large roads (even highways). You can access them in the road creation tool at the bottom of the screen, next to the electricity lightning bolt icon. Pick the type of one-way road you want to make and just lay it down whenever you're ready to place it. Make sure the road connect to everything in a logical way, to ensure drivers actually use it.

What are one-way roads used for in Cities Skylines 2?

One-way roads are best used to relieve traffic congestion in areas where lots of vehicle traffic is heading in one direction, such as highways leading between residential and commercial zones. You can turn one-way roads into these sort of express-ways to get some cars to leave the busier road and reduce overall traffic.
