The yellow star marks a door that will not open if you kill any civilians. Civilians will appear red if you place your cursor over them but will not aggro on you unless you attack them. You will need to aggro the cultists around them and lead them away before killing them. Three seeds are behind this door.

The white star marks a door that will not open unless you kill all cultists and demons. Make sure to check every room as small groups can be missed easily. As long as you don't encounter the boss, you can backtrack to look for missed cultists. Two seeds are behind this door.
